    Application Example



    Figure 1: PCR amplification using EURx Perpetual OptiTaq DNA Polymerase.
    A 2.5 kb PCR amplicon was amplified with Perpetual OptiTaq DNA Polymerase either in stand-alone or in master mix format, respectively.
    • Lane MW (1): molecular size marker- Perfect 1 kb DNA Ladder (Cat. No. E3130).
    • Lanes "Separate Components" (2,3): PCR amplification reactions using 1.25 U Perpetual OptiTaq DNA Polymerase, Pol Buffer B and dNTPs.
    • Lanes "Master Mix" (4,5): PCR amplification reactions using Perpetual OptiTaq PCR Master Mix (2x), after 25 freeze-thaw cycles.
    • Lanes "Master Mix + Dye" (6,7): PCR amplification reactions using Perpetual OptiTaq PCR Master Mix (2x) and 10 x Color Load, after 25 freeze-thaw cycles.
    Result: Following 25 freeze-thaw cycles, OptiTaq Master Mix still displayed similar performance to OptiTaq DNA Polymerase with separately stored components and not subjected to repeated freeze-thawing.

Lanes 1.1 to 20 kb: PCR amplification reactions (each reaction 1.25 U EURx Perpetual OptiTaq DNA Polymerase, Pol Buffer B, 0.2 mM dNTPs; 50 µl reaction volume).
