Application Example
Figure: Sensitive HotStart PCR amplification with Perpetual Taq DNA Polymerase (Cat. No. E2700) in a size range between ~1 and 15 kb. Molecular weight marker M1 (Perfect™ 1kb DNA Ladder). Lanes 1.1 to 15 kb: PCR amplification reactions (each reaction 1.25 U EURx Perpetual Taq DNA Polymerase, Pol Buffer B, 0.2 mM dNTPs; 50 µl reaction volume).
Notes:
Color Perpetual Taq DNA Polymerase is supplemented with two tracing dyes (yellow and red). These dyes allow direct loading of PCR products to agarose gels. Additionally, these dyes provide an optical control for a complete mixing during reaction setup by visualizing the complete suspension of polymerase in the reaction mix. This may prove useful for less experienced users such as teaching classes and student courses. In this Color edition, these dyes are mixed directly to the polymerase . As a consequence, the Color enzyme edition does not allow the end user to run PCR reactions without addition of dyes.
Note, that uncoloured standard Perpetual Taq DNA Polymerase contains both dyes in a separate coloured buffer, giving you a choice to run both colored and uncolored PCR reactions. Thus, when looking for a 10x buffer / optional gel loading dye combination, experienced researchers will want to use Standard Perpetual Taq DNA Polymerase . Among colourless buffers, the standard enzyme package contains a purple coloured buffer Pol C. Purple buffer Pol C allows to load aliquots of PCR reactions directly on gel without the need to add a separate gel loading buffer.
